SUMMARY
The incubator model Ninc-S60 is designed for incubating NRI and NSC biological indicators for Steam sterilization. This model is set on a temperature 55-60°C.There are 15 incubation slots available to place the self-contained biological indicators.For convenience a crushing well is integrated to enable the user to activate the BI’s.The Etigam NRI/NSC consists of a self-contained unit that includes bacterial spores inoculated onto a paper filter carrier and a small glass ampoule containing a culture medium all encased in a plastic vial that serves as the culture tube.After removing the NRI/NSC from the sterilizer and confirming that the chemical indicator printed on the label of the NSC has turned brown/black, the NRI/NSC needs to be activated using the integrated crushing well on the device or a separate vial crusher. Assure that the growth medium has saturated the spore carrier inside the tube. The now activated NRI/NSC needs to be incubated according to the specifications and regularly checked for spore growth. Spore growth can be noticed by color change of the medium and/or turbidity in the tube. If the indicator turns yellow, the test is positive indicating inadequate sterilization.
SAFETY
Install according to specifications on the incubator and in conformance with local electricity codes. PREPARATION
1. Remove all packaging from the incubator and inspect for damage.
2. Plug into your local earthed and grounded power source and green light will turn on.
3. Allow incubator to warm up during 30 minutes.The incubator is designed to work continuously and does not have a power switch.
4. Refer to the Technical Data Sheet of the NRI/NSC for complete instructions.
OPERATION
1. Check if the green light in the front of the incubator is illuminated.
2. Use a thermometer (manual of digital) to check periodically if the temperature of the wells of the incubator are within the range 55-60°C.
3. Activate the processed NSC by using the integrated crushing well on the incubator or a separate vial crusher and place immediately in the incubation slots recording incubation start time and slot number.
4. Place the unprocessed positive control (from the same lot number) in an incubation slot as well recording start time and slot number.
5. Incubate at 55-60°C for 10 hrs for the NRI and 24 hrs for the NSC checking for spore growth (visual color change from purple to yellow and/or turbidity) at regular intervals.
6. The positive control should change color from purple to yellow within 10 hrs for the NRI and 24 hrs for the NSC.
7. Use the Technical Data Sheet of the NRI and NSC for further interpretation.
CLEANING
1. Before cleaning, unplug the incubator from the power source and allow to cool down.
2. Use a cloth with isopropyl alcohol for the exterior.
3. Use a cotton swab dampened with isopropyl alcohol for the incubation slots and crushing well.
